Professional Statement

Lysosomes maintain neuronal proteostasis by degrading aggregated proteins such as tau. In Alzheimer’s disease (AD), lysosomal dysfunction impairs this process, leading to tau accumulation and neurofibrillary tangles. We hypothesize that intracellular tau aggregation disrupts lysosomal composition and function, further promoting pathology. To test this, we will characterize changes in lysosomal proteins and function under tau aggregation conditions using tau biosensor HEK293 cells and human iPSC-derived neurons from AD patients. Key findings will be validated in mouse models. This work will define how tau impairs lysosomal function and identify potential therapeutic targets for restoring proteostasis in AD.
